结肠癌组织中MEG8、TGM2 mRNA的表达变化及其意义Expression and significance of MEG8 and TGM2 mRNA in colon cancer tissues

摘要(Abstract):
目的观察结肠癌组织中长链非编码RNA(lncRNA)母源性表达基因8(MEG8)、转谷氨酰胺酶2(TGM2)mRNA的表达变化,并探讨其意义。方法采用实时荧光定量PCR法检测85例份结肠癌组织及其癌旁正常组织MEG8、TGM2 mRNA,分析两者间及其与结肠癌临床病理参数、患者预后的关系。结果结肠癌及其癌旁正常组织MEG8 mRNA相对表达量分别为0.62±0.23、1.87±0.55,TGM2 mRNA相对表达量分别为2.03±0.95、0.98±0.36,两者比较,P均<0.05。MEG8、TGM2 mRNA表达均与结肠癌TNM分期、淋巴结转移有关(P均<0.05)。结肠癌组织中MEG8 mRNA和TGM2 mRNA表达呈显著负相关(r=-0.798,P<0.05)。MEG8 mRNA高表达者5年总生存率为77.78%(35/45)、生存时间为(43.62±8.31)个月,低表达者分别为52.50%(21/40)、(31.07±7.25)个月,两者比较,P均<0.05;TGM2 mRNA高表达者5年总生存率为54.55%(24/44)、生存时间为(37.21±9.68)个月,低表达者分别为78.49%(32/41)、(49.18±8.76)个月,两者比较,P均<0.05。结肠癌患者预后Cox回归模型分析显示,高级别TNM分期、有淋巴结转移、MEG8 mRNA低表达、TGM2 mRNA高表达是影响结肠癌患者预后的危险因素(P均<0.05)。结论结肠癌组织中MEG8 mRNA呈低表达,TGM2 mRNA呈高表达,两者表达均与TNM分期、淋巴结转移有关,检测两指标有助于评估疾病病情及患者预后。
