Abstract:Objective To investigate research hotspots regarding lipogenesis mechanisms in hepatocytes in metabolic dysfunction-associated steatotic liver disease (MASLD) and their molecular regulation. Methods A search of the Web of Science Core Collection was conducted for literature related to lipogenesis published from January 1, 2011 to September 1, 2025. Bibliometric analysis and visualization were performed using VOSviewer 1.6.20 and CiteSpace 6.3.R1. Results A total of 635 English-language articles were included. The annual publication output showed an overall upward trend, with a particularly marked increase after 2016; the highest annual output was 7.4 times that of 2011. China and the United States ranked first and second in publication volume; the United States led in total citations (14,592) and in the number of high-impact institutions. Journals with high publication output all fell within the JCR Q1/Q2 categories, with Nutrients publishing the most articles and Hepatology receiving the highest citations. Core author Hodson L systematically reviewed the role of lipogenesis in metabolic diseases; the most highly cited article was the pioneering study by Donnelly K L, which was the first to quantitatively determine the sources of hepatic fat. Co-citation cluster analysis revealed that the regulation of lipogenesis, interaction with insulin resistance (IR), and dietary factors were the current research focuses. Keyword analysis showed that lipogenesis and IR were the two central research hotspots in this field. In addition, fructose metabolism, AMP-activated protein kinases (AMPK) regulation, and disorders of cholesterol metabolism were gradually emerging as new research directions. Conclusion Research on the regulation of lipid synthesis in MASLD shows significant potential. The United States and China are the major contributing countries, and Hodson L and Donnelly KL are the core influential authors. Lipid regulation and IR are the current research hotspots, while fructose metabolism, AMPK signaling, and disorders of cholesterol metabolism are emerging frontiers. Bibliometric and visualization analysis can effectively sort out the research context, and help track frontiers and predict trends.
