子宫内膜癌组织中NORAD、FUBP1 mRNA表达变化及其意义

摘要:

目的 观察子宫内膜癌(EC)组织中DNA损伤激活的非编码RNA(NORAD)、远端上游元件结合蛋白1(FUBP1)mRNA的表达变化,并探讨其临床意义。方法 接受手术治疗的EC患者138例,术中留取EC癌组织和癌旁组织,采用qRT-PCR法检测EC癌组织和癌旁组织中NORAD、FUBP1 mRNA及凋亡相关基因Bax、p53、Bcl-2 mRNA,分析EC癌组织中NORAD、FUBP1 mRNA表达的相关性及其与凋亡相关基因Bax、p53、Bcl-2 mRNA表达的相关性,并分析NORAD、FUBP1 mRNA表达与EC患者临床病理参数的关系。患者出院后进行随访,随访至2025年7月,计算其3年总体生存(OS)率和3年总无进展生存(PFS)率,以NORAD、FUBP1 mRNA相对表达量的平均值为临界值,将138例EC患者分为NORAD高表达者、NORAD低表达者和FUBP1 mRNA高表达者、FUBP1 mRNA低表达者,并分析NORAD、FUBP1 mRNA相对表达量与EC患者预后的关系。结果 EC癌组织中NORAD、FUBP1 mRNA相对表达量分别为0.82±0.20、3.11±0.63,EC癌旁组织中NORAD、FUBP1 mRNA相对表达量分别为3.22±0.56、0.74±0.22,两者相比,P均<0.05。EC癌组织中NORAD与FUBP1 mRNA表达呈负相关关系(r=-0.718,P<0.001)。EC癌组织中Bax、p53 mRNA相对表达量低于癌旁组织,Bcl-2 mRNA相对表达量高于癌旁组织(P均<0.05)。EC癌组织中NORAD与Bax、p53 mRNA表达呈正相关关系(r分别为0.662、0.651,P均<0.001),与Bcl-2 mRNA表达呈负相关关系(r=-0.712,P<0.001);EC癌组织中FUBP1 mRNA与Bax、p53 mRNA表达呈负相关关系(r分别为-0.715、-0.704,P均<0.001),与Bcl-2 mRNA表达呈正相关关系(r=0.656,P<0.001)。NORAD、FUBP1 mRNA表达与EC患者FIGO分期、淋巴结转移有关(P均<0.05)。NORAD高表达者3年PFS率高于NORAD低表达者(P<0.05),FUBP1 mRNA高表达者3年PFS率低于FUBP1 mRNA低表达者(P<0.05)。结论 EC癌组织中NORAD表达下调、FUBP1 mRNA表达上调,两者呈负相关关系。EC癌组织中NORAD、FUBP1 mRNA的表达与凋亡相关基因的表达相关,也与EC患者FIGO分期、淋巴结转移有关。NORAD低表达、FUBP1 mRNA高表达的EC患者预后较差。

Abstract:

Objective To observe the expression changes of non-coding RNA activated by DNA damage(NORAD) and far-upstream element binding protein 1(FUBP1) mRNA in the endometrial carcinoma(EC) tissues, and to explore their clinical significance. Methods A total of 138 EC patients who underwent surgical treatment were enrolled. EC cancer tissues and adjacent non-cancerous tissues were collected during surgery. The qRT-PCR was used to detect the expression of NORAD, FUBP1 mRNA, as well as apoptosis-related genes Bax, p53, and Bcl-2 mRNA in EC cancer tissues and adjacent non-cancerous tissues. The correlation between the expression of NORAD and FUBP1 mRNA in EC cancer tissues, as well as their correlation with the expression of apoptosis-related genes Bax, p53, and Bcl-2 mRNA, was analyzed. Furthermore, the relationship between the expression of NORAD and FUBP1 mRNA and the clinicopathological parameters of EC patients was investigated. After discharge, patients were followed up until July 2025. The 3-year overall survival(OS) rate and 3-year progression-free survival(PFS) rate were calculated. The average relative expression levels of NORAD and FUBP1 mRNA were used as the cut-off value, the 138 EC patients were divided into the high and low NORAD expression groups, as well as the high and low FUBP1 mRNA expression groups. The relationships between the relative expression levels of NORAD and FUBP1 mRNA and the prognosis of EC patients were analyzed. Results The relative expression levels of NORAD and FUBP1 mRNA in EC cancer tissues were 0.82 ± 0.20 and 3.11 ± 0.63, respectively; the relative expression levels of NORAD and FUBP1 mRNA in adjacent non-cancerous tissues were 3.22 ± 0.56 and 0.74 ± 0.22, respectively; there were significant differences between these two kinds of tissues(all P<0.05). In EC cancer tissues, there was a negative correlation between the expression of NORAD and FUBP1 mRNA(r=-0.718, P<0.001). The relative expression levels of Bax and p53 mRNA in EC cancer tissues were lower than those in adjacent non-cancerous tissues, while the relative expression level of Bcl-2 mRNA was higher(all P<0.05). In EC cancer tissues, there was a positive correlation between NORAD and the expression of Bax and p53 mRNA(r=0.662 and 0.651, respectively, both P<0.001), and a negative correlation with the expression of Bcl-2 mRNA(r=-0.712, P<0.001). In EC cancer tissues, FUBP1 mRNA expression was negatively correlated with the expression of Bax and p53 mRNA(r=-0.715 and-0.704, respectively, both P<0.001), and was positively correlated with the expression of Bcl-2 mRNA(r=0.656, P<0.001). The expression of NORAD and FUBP1 mRNA was associated with FIGO staging and lymph node metastasis in EC patients(both P<0.05). The 3-year PFS rate was higher in patients with high NORAD expression than in those with low NORAD expression(P<0.05), and the 3-year PFS rate was lower in patients with high FUBP1 mRNA expression than in those with low FUBP1 mRNA expression(P<0.05). Conclusions In EC cancer tissues, NORAD expression is down-regulated, FUBP1 mRNA expression is up-regulated, and there is a negative correlation between them. The expression of NORAD and FUBP1 mRNA in EC cancer tissues is correlated with the expression of apoptosis-related genes, as well as FIGO staging and lymph node metastasis in EC patients. EC patients with low NORAD expression and high FUBP1 mRNA expression have a poor prognosis.
